Still trying to find a ChIP antibody that works?
The HaloCHIP™ System is a novel method that covalently captures intracellular protein:DNA complexes without using antibodies.
In the HaloCHIP™ method, a HaloTag® fusion protein is expressed either transiently or stably in mammalian cells and crosslinked complexes can be directly captured from a cellular lysate via covalent binding to a HaloTag®-specific resin, termed HaloLink™ resin. The covalent linkage established at this point allows for extensive washing to remove non-specific protein and DNA, followed by standard reversal of the crosslinks to release the DNA fragments bound to the DNA-binding protein.
- Simple: 1 day ChIP protocol.
- Low Background: HaloTag® covalent capture allows stringent washing compared to antibody-dependent ChIP.
- Value: Savings over conventional ChIP kits which require the purchase of an untested antibody.

qPCR Data.Fold enrichment patterns are maintained using the HaloCHIP™ system to pulldown CREB:DNA partners compared to conventional CREB ChIP.
ChIP-chip data. High-resolution analysis demonstrates nearly identical promoter enrichment profiles for CREB binding when comparing CREB-HaloCHIP™ to CREB-ChIP.
