GPCRs play an important role in a variety of biological processes, including immune system activity and behavior regulation. GPCRs are popular targets in drug discovery research and a well-designed assay can help expedite the discovery of novel candidates. Many pharmaceutical high-throughput screening (HTS) programs desire a simple, sensitive, automatable GPCR-specific cell-based assay with a wide dynamic range and low susceptibility to compound interference. In this poster we present the new Promega GloSensor™ cAMP Assay, an HTS-friendly bioluminescent assay capable of measuring intracellular cAMP levels in living cells in real-time. Two HTS instruments are used for dispensing components of this application to screen for effects on GPCRs. Specifically, ultra low volumes of compounds are dispensed into 1536-well assay plates using the Labcyte® Echo® 555 acoustic liquid handler, and GloSensor™ cAMP cells are dispensed using the Deerac HTS reagent dispense. Implementing a miniaturized GPCR assay as demonstrated in this poster allows cost-effective screening for modulators of GPCR function, a useful tool for labs in the pharmaceutical HTS industry.
